Myself and a few friends from Zurich went on a quick day trip to Stein am Rhein this weekend. My friends have a car so we made the 45/50 minute drive around 1pm on a sunny Sunday afternoon, leaving around 6pm for the drive back to Zurich.
This amount of time allowed us to wander the town, poke into shops and stop for coffee and cake (or beer) along the waterfront. We also made a quick trip up to Hohenklingen Castle (5 minute car ride otherwise a 30/45 minute hike up the hill (I'm guessing). There was a nice looking restaurant there which had great views of the town and you might spend 45 to 60 minutes exploring the castle (it isn't too big).
There were also boat cruises running when we were there (April 23).
If you take a train (which my wife and I would do normally as we don't own a car) you can get there in about 1 hour from Zurich main train station - schedules here. It is a very small town and is easy to explore on foot in one afternoon.
Also we were pleasantly surprised at food and drink prices - due we're guessing to their desire to attract nearby Germans for the afternoon.
