Yes, you need both a reservation and a valid rail pass or ticket for the Glacier Express. This page explain how it works: https://www.myswissalps.com/glacierexpress/tickets. You'll also find links to trustworthy resellers at that page.
Before you buy any ticket: will you be making other journeys in Switzerland too? And if so, will you be using a rail pass (https://www.myswissalps.com/train/ticketspasses)? Some rail passes, such as the Swiss Travel Pass (https://www.myswissalps.com/swisstravelpass), fully cover the Glacier Express journey. In that case all you need are seat reservations, but no additional tickets.
If you don't have a rail pass, a Saver Day Pass (https://www.myswissalps.com/saverdaypass) can be cheaper than normal tickets (https://www.myswissalps.com/traintickets/switzerland) for your Glacier Express trip.
