Yes, reservations on the Bernina Express bus between Lugano and Tirano are mandatory. As this service only runs once per day in both directions, seats are limited, so don’t leave it too late to book.
If you travel on regional trains instead of the Bernina Express train between Tirano and Chur, reservations are not needed (or possible).

There are no other buses between Lugano and Tirano apart from the Bernina Express bus. Other options for getting between Lugano and Tirano are rather convoluted and take much more time.
If you don’t want to go via Tirano, there are two options for travelling between Lugano and Chur utilising PostBus services, which are covered by the Swiss Travel Pass: the Palm Express PostBus to St Moritz and the PostBus from Bellinzona over the San Bernardino Pass to Chur. Both those options also involve some train sectors, which are also coverd by the Swiss Travel Pass. The Palm Express is a seasonal service and not available every day of the year. Reservations are compulsory (and free) on the Palm Express PostBus and the PostBus from Bellinzona to Chur.

Thanx for the guidance. Pls tell me if the reservations for BEX bus from lugano to tirano can be done on SBB or it has to be done through RHB site only.

You can make reservations through the Rhätische Bahn (Rhaetian Railway) site. If you are travelling in June, the cost is CHF 14 per seat.
