Hi Deni,
The Glacier Express (route) is just a more scenic way of getting to Chur from Lucerne then the standard Lucerne to Zurich to Chur route. You can take the actual Glacier Express or regular trains following the same route.
The boat trip to Flüelen is wonderful.
Andermatt to Chur is only 2h 21m. The latest GEX (as the Glacier Express is denoted on the timetable) departs Andermatt 12:54. and arrives Chur at 15:15. Chur has a nice old town but there is not much else to see there. I'd say you could walk right round it in a couple of hours.
You could of course spend the morning in Lucerne visiting Rigi or Pilatus or Titlis even and still make it to Chur at a reasonable hour. The fastest connection is just over 2 hours via Thalwil near Zurich.
If you are at Lauterbrunnen a very nice circuit to do is to Grütschalp (cable car), Mürren (cogwheel train), Gimmelwald (cable car), Stechelberg (cable car), Lauterbrunnen (bus). From Mürren you can easily visit Schiltorn if you have time. In between Stechelberg and Lauterbrunnen the bus stops at the Trummelbach Falls. Well worth a look.

PS Out of interest I had a look at the Swiss Timetable for February. There is a train leaving Tirano at 13:08 taking 5h 50m to reach Castlefranco Veneto with 2 changes. Connections are not yet showing for June but should be pretty similar. See attached.
